The U.S. Department of Agriculture–Agricultural Research Service (USDA-ARS) has been a leader in weed science research covering topics ranging from the development and use of integrated weed management (IWM) tactics to basic mechanistic studies, including biotic resistance of desirable plant communities and herbicide resistance. ARS weed scientists have worked in agricultural and natural ecosystems, including agronomic and horticultural crops, pastures, forests, wild lands, aquatic habitats, wetlands, and riparian areas. Through strong partnerships with academia, state agencies, private industry, and numerous federal programs, ARS weed scientists have made contributions to discoveries in the newest fields of robotics and genetics, as well as the traditional and fundamental subjects of weed–crop competition and physiology and integration of weed control tactics and practices. Weed science at ARS is often overshadowed by other research topics; thus, few are aware of the long history of ARS weed science and its important contributions. This review is the result of a symposium held at the Weed Science Society of America’s 62nd Annual Meeting in 2022 that included 10 separate presentations in a virtual Weed Science Webinar Series. The overarching themes of management tactics (IWM, biological control, and automation), basic mechanisms (competition, invasive plant genetics, and herbicide resistance), and ecosystem impacts (invasive plant spread, climate change, conservation, and restoration) represent core ARS weed science research that is dynamic and efficacious and has been a significant component of the agency’s national and international efforts. This review highlights current studies and future directions that exemplify the science and collaborative relationships both within and outside ARS. Given the constraints of weeds and invasive plants on all aspects of food, feed, and fiber systems, there is an acknowledged need to face new challenges, including agriculture and natural resources sustainability, economic resilience and reliability, and societal health and well-being.
Inflammation throughout pregnancy and fetal growth restriction in rural Nepal

Maternal systemic inflammation during pregnancy may restrict embryo−fetal growth, but the extent of this effect remains poorly established in undernourished populations. In a cohort of 653 maternal−newborn dyads participating in a multi-armed, micronutrient supplementation trial in southern Nepal, we investigated associations between maternal inflammation, assessed by serum α1-acid glycoprotein and C-reactive protein, in the first and third trimesters of pregnancy, and newborn weight, length and head and chest circumferences. Median (IQR) maternal concentrations in α1-acid glycoprotein and C-reactive protein in the first and third trimesters were 0.65 (0.53–0.76) and 0.40 (0.33–0.50) g/l, and 0.56 (0.25–1.54) and 1.07 (0.43–2.32) mg/l, respectively. α1-acid glycoprotein was inversely associated with birth size: weight, length, head circumference and chest circumference were lower by 116 g (P = 2.3 × 10−6), and 0.45 (P = 3.1 × 10−5), 0.18 (P = 0.0191) and 0.48 (P = 1.7 × 10−7) cm, respectively, per 50% increase in α1-acid glycoprotein averaged across both trimesters. Adjustment for maternal age, parity, gestational age, nutritional and socio-economic status and daily micronutrient supplementation failed to alter any association. Serum C-reactive protein concentration was largely unassociated with newborn size. In rural Nepal, birth size was inversely associated with low-grade, chronic inflammation during pregnancy as indicated by serum α1-acid glycoprotein.

OBJECTIVES/SPECIFIC AIMS: Recent evidence from resting-state fMRI studies have shown that brain network connectivity is altered in patients with neurodegenerative disorders. However, few studies have examined the complete connectivity patterns of these well-reported RSNs using a whole brain approach and how they compare between dementias. Here, we used advanced connectomic approaches to examine the connectivity of RSNs in Alzheimer disease (AD), Frontotemporal dementia (FTD), and age-matched control participants. METHODS/STUDY POPULATION: In total, 44 participants [27 controls (66.4±7.6 years), 13 AD (68.5.63±13.9 years), 4 FTD (59.575±12.2 years)] from an ongoing study at Indiana University School of Medicine were used. Resting-state fMRI data was processed using an in-house pipeline modeled after Power et al. (2014). Images were parcellated into 278 regions of interest (ROI) based on Shen et al. (2013). Connectivity between each ROI pair was described by Pearson correlation coefficient. Brain regions were grouped into 7 canonical RSNs as described by Yeo et al. (2015). Pearson correlation values were then averaged across pairs of ROIs in each network and averaged across individuals in each group. These values were used to determine relative expression of FC in each RSN (intranetwork) and create RSN profiles for each group. RESULTS/ANTICIPATED RESULTS: Our findings support previous literature which shows that limbic networks are disrupted in FTLD participants compared with AD and age-matched controls. In addition, interactions between different RSNs was also examined and a significant difference between controls and AD subjects was found between FP and DMN RSNs. Similarly, previous literature has reported a disruption between executive (frontoparietal) network and default mode network in AD compared with controls. DISCUSSION/SIGNIFICANCE OF IMPACT: Our approach allows us to create profiles that could help compare intranetwork FC in different neurodegenerative diseases. Future work with expanded samples will help us to draw more substantial conclusions regarding differences, if any, in the connectivity patterns between RSNs in various neurodegenerative diseases.

We conducted a longitudinal study involving 734 college students over a three-month period that included the 2008 U.S. presidential election. The study investigated factors such as respondents' personality characteristics and ideological proclivities in predicting perceptions of the major candidates and both stability and change in voting preferences for Barack Obama and John McCain. Previous research on personality and political orientation suggests that Openness to New Experiences is positively associated with liberal political preferences, whereas Conscientiousness is positively associated with conservative preferences; we replicated these results in the context of the current study. Several ideological factors also predicted conversion to Obama's candidacy. These included respondents' degree of self-reported liberalism, perceptions of their parents as liberal (versus conservative), and lower scores on measures of authoritarianism and political system justification (i.e., support for the prevailing system of electoral politics and government). The effects of Openness and Conscientiousness on candidate preferences were statistically mediated by ideological variables, providing further evidence that general predispositions exist that link personality and political orientation, and these are likely to play a significant role in electoral politics. Implications for the integration of “top-down” (institutional) and “bottom-up” (psychological) approaches to the study of political behavior are discussed.

Examination of the eyes with a slit lamp revealed that 101/H mice had a coloured cataract. Crosses to C3H/HeH indicated that this was inherited as a single recessive gene which we have designated lop-2 (lens opacity-2). The related strain 129/Sv-SlJ-CP had a phenotypically identical cataract and presumably also carries the lop-2 gene. CBA/H, CBA/CaH- + /p, CBA/H-kd and CBA/H-T6 mice had a bright white or white/green cataract that typically extended from the nucleus to the anterior cortex of the lens. Crosses to C3H/HeH indicated that this was inherited as a semi-dominant gene. However, other crosses raise the possibility that the CBA cataract is also caused by lop-2. If so, the expressivity (and penetrance of the heterozygote) is affected by genetic background. Neither lop-2 nor the gene responsible for the CBA cataract was linked to contrasted (Slcon) on chromosome 10, so these are distinct from the Lop (lens opacity) gene. Further studies of genetic linkage are needed to clarify whether lop-2 is responsible for both the 101/H and CBA/H cataracts.

Two series of mouse chimaeras were produced by aggregating pairs of eight-cell embryos that differed at the Gpi-1s locus, encoding glucose phosphate isomerase (GPI-1); the paired embryos were respectively homozygous Gpi-1sa/Gpi-1sa and Gpi-1sb/Gpi-1sb. Chimaeric blastocysts were transferred to pseudopregnant females, that were homozygous Gpi-1se/Gpi-1se and produced only GPI-1 C enzyme. Quantitative electrophoresis of GPI-1 was used to estimate the contribution of each embryo (GPI-1 A and GPI-1B enzyme activity) to the foetus, placenta and other extraembryonic tissues of 12½ day chimaericconceptuses. For both series of chimaeras, the distributions of %GPI-1A in different tissues were classified as (1) balanced and typical, (2) balanced but atypical or (3) unbalanced. One series of chimaeras was clearly unbalanced, so that the cells derived from the (C57BL × CBA/Ca)F2 embryo (Gpi-1sb/Gpi-1sb) predominated over those derived from the BALB/c inbred strain (Gpi-1sa/Gpi-1sa) in most foetuses. Two significant observations were made concerning this unbalanced series. Firstly, the mean composition of the placenta and other extraembryonic tissues was similar to that in the foetus i.e. also unbalanced with (C57BL × CBA/Ca)F2 (abbreviated to BF2) cells predominating. Secondly, despite this generalizeddeficiency of BALB/c cells, there were differences in the frequency of non-chimaeric tissues between different developmental lineages. In 20/34 chimaeric conceptuses in the unbalanced series only BF2 cells were detected in the foetus, whereas both BF2 and BALB/c cells were present in at least one of the extraembryonic tissues. This group of chimaeras, therefore, shows some similarities to human confined mosaicism. Although chimaerism occurred more often in the primitive endoderm (hypoblast) lineage (yolk sac endoderm and parietal endoderm) than in the placenta, this may also be the case in human mosaics. The mosaic status of the human yolk sac endoderm is usually unknown so it is possible that mosaicism often occurs in the yolk sac endoderm as well as the trophectoderm in human ‘confined placental mosaicism’. The uniformly unbalanced phenotype seen in the mouse chimaeras may be a result of generalized cell selection against BALB/c cells in all tissues. As an alternative explanation, we propose that most of the BALB/c cells in the blastocyst are allocated to the mural trophectoderm, which has a limited mitotic potential and so contributes little to the mid-gestation conceptus. Further work is required to test these possibilities.

The electrophoretic variants of the X-chromosome-linked enzyme phosphoglycerate kinase (PGK-1) have been used to investigate the randomness of X chromosome expression in the fetus and various extra-embryonic membranes of the mouse conceptus. The amnion shows essentially random expression of the maternally derived X chromosome (Xm) and the paternally derived X chromosome (Xp). The parietal endoderm, however, shows exclusive or preferential expression of Xm. The results support the idea that the randomness of X chromosome expression is correlated with embryonic cell lineage such that Xm is preferentially (perhaps exclusively) expressed in derivatives of the primitive endoderm and trophectoderm but that Xm and Xp are randomly expressed in the derivatives of the primitive ectoderm.
Experiments involving ovary transplants, embryo transfers or crosses with heterozygous mothers confirm previous findings that Xm is preferentially expressed regardless of the X chromosome expressed in the reproductive tract. Additional experiments show that the preferentially expressed X chromosome in the parietal endoderm and visceral yolk sac endoderm of a normal XmXp conceptus is always Xm regardless of grand-parental origin of Xm and regardless of whether the mother is a normal XX female or an XO female. Xp is, however, expressed in these tissues hi XpO female conceptuses. It is argued that a form of chromosome imprinting occurs at each generation to mark Xm and Xp as different and that this difference influences the choice of which X chromosomes are expressed in each cell lineage.
Dominant hemimelia and En-1 on mouse chromosome 1 are not allelic

Previous studies have shown that En-1, a homeobox-containing gene, maps close to or at the Dh locus in the mouse. Since homeobox-containing genes are key genes in the control of development the close proximity of En-1 to the developmentally significant gene Dh raised the possibility that the Dh mutation represented a mutant allele of En-1. A genetic analysis involving En-1, Dh, and other chromosome 1 markers (Emv-17, In and Pep-3) shows that although Dh and En-1 are closely linked they are separable by recombination (4/563). The likely gene order and recombination frequencies of these loci are: In (5.2±0.9) Emv-17 (1.1±0.4) Dh (0.7±0.4) En-1 (3.0±0.7) Pep-3. This shows that Dh is not a mutant allele of En-1.

Previous studies have demonstrated that the LT/SvKau strain of mice ovulates a high proportion of oocytes as diploid primary oocytes rather than secondary oocytes. These ovulated primary oocytes are arrested at meiotic metaphase I but may be fertilized to produce digynic triploid embryos. In the present study, 40·4% of eggs analysed from LT/SvKau females were ovulated as primary oocytes, compared to 1·2% from control C57BL/Ws strain mothers. These two inbred strains were intercrossed to produce eight sets of Fl, F2 and backcross females and the frequency of triploidy was investigated. The results are compatible with segregation of a co-dominant, autosomal gene that has a major influence on the incidence of triploidy. We suggest that the provisional gene symbol Poo (primary oocyte ovulation) be assigned to this gene, with alleles Pool (the ‘mutant’ allele present in the LT/SvKau strain) and Poob (the normal allele present in C57BL/Ws mice). Poo is incompletely penetrant and has variable expressivity because the proportion of oocytes ovulated as primary oocytes by LT/SvKau mice was variable and, in some cases, nil. In putative Pool/Poob heterozygotes the frequency of ovulated primary oocytes was increased only marginally (from 1·2% to 66%) by the presence of one copy of the Pool allele, but this increase was found consistently (in two reciprocal Fl crosses) and was statistically significant. No evidence was found for tight genetic linkage between Poo and two Mendelian loci (brown on chromosome 4 and glucose phosphate isomerase on chromosome 7), that were segregating in the crosses. The Pool mutant in the LT/SvKau strain of mice provides a valuable resource to study the cell and molecular biology of mammalian oocyte maturation and the control of meiosis.

The effect of the Xce (x-chromosome controlling element) genotype on the randomness of X-chromosome inactivation in the mouse was studied by monitoring the expression of the X-linked locus pgk-1. The main aim was to test whether the Xce genotype modified the preferential expression of the maternally derived X-chromosome in the yolk sac endoderm. Quantitative electrophoresis of phosphoglycerate kinase (PGK-1) was used to study Pgk-1 expression in the foetus, yolk sac mesoderm and yolk sac endoderm at 13½ days post coitum. The Xcea/Xcec genotype caused non-random X-chromosome expression in the foetus and yolk sac mesoderm. However, there was no evidence that the Xce genotype moderates the preferential expression of the maternally derived X-chromosome in the yolk sac endoderm, as reported by Rastan & Cattanach (1983).

Two substrains of the inbred mouse strain 101, maintained at Harwell and at Neuherberg, and designated 101/H and 101/E1, differed at five of the eight genetic loci tested and it seems very likely that one of these substrains has been genetically contaminated. The available evidence suggests that 101/E1 is probably the contaminated substrain.

The dimeric enzyme glucose phosphate isomerase (GPI-1) is regulated in oocytes by a cis-acting temporal gene (Gpi-1t) that maps close to the structural gene (Gpi-1s). Quantitative cellulose acetate electrophoresis of GPI-1 allozymes from unfertilized eggs produced by various Gpi-1sa / Gpi-1sb heterozygous females revealed a new Gpi-1t allele that we have designated Gpi-1tc. This allele is present in 101/H mice and a partially congenie stock that carries the Gpi-lsa gene derived from the AKR strain. We have confirmed that Gpi-1tc is closely linked to Gpi-1s and that it is cis-acting. It produces higher levels of GPI-1 in unfertilized eggs than the other two Gpi-lt alleles that are known (Gpi-1ta and Gpi-1tb) but has no effect on GPI-1 in somatic tissues or spermatozoa. This new Gpi-1t allele represents a third developmental programme for GPI-1 expression in oocytes.

Two series of 12½ day mouse chimaeric conceptuses were produced by aggregating (C57BL × CBA)F2 strain preimplantation embryos with embryos that differed at the Gpi-1s locus that encodes glucose phosphate isomerase, GPI-1. The composition of individual issues was evaluated by quantitative electrophoresis to estimate the % GPI-1A in the chimaeric tissue containing GPI-1A and GPI-1B. In one series of chimaeras, the GPI-1A cells were derived from a backcross between inbred BALB/c strain females and (BC × BALB/c)F1 males, where BC is the partly congenic strain C57BL/Ola.AKR-Gpi-lsa,c/Ws. In the other series of chimaeras, the GPI-1A cells were derived from the reciprocal backcross between (BC × BALB/c)F1 females and inbred BALB/c strain males. The [(BC × BALB/c)F1 female × BALB/c male] ↔ (C57BL × CBA)F2 series of chimaeras was reasonably balanced so that GPI-1 A and GPI-1B cells were fairly equally represented in the foetuses, placentas and extraembryonic membranes (tissue means: 37–51 % GPI-1A). This series did not differ significantly in composition from an earlier series of (BC × BALB/c)F2 ↔ (C57BL × CBA)F2 chimaeras. However, the [BALB/c female × (BC × BALB/c)F1 male] ↔ (C57BL × CBA)F2 series of chimaeras was unbalanced, with mean tissue compositions (28–33% GPI-1A) that were intermediate between the above two balanced series and the unbalanced (BALB/c × BALB/c) ↔ (C57BL × CBA)F2 series (tissue means: 14–22% GPI-1 A), that was studied previously. Thus, both (BALB/c×BALB/c) and [BALB/c×(BC x BALB/c)F1 embryos contributed less to the tissues of chimaeric conceptuses than either (BC × BALB/c)F2or [(BC × BALB/c)F1 × BALB/c] embryos. This implies that embryos from BALB/c mothers contributed less to the tissues of chimaeric conceptuses than embryos from (BC × BALB/c)F1 mothers. We, therefore, conclude that a maternal genetic effect is responsible for some of the differences in composition among the four groups of chimaeras. This maternal effect must act before the 8-cell stage but it is not yet known whether it is mediated via cytoplasmic inheritance, genomic imprinting or by the reproductive tract. Evidence that a maternal effect retards preimplantation development of embryos from BALB/c females is reviewed and the possibility that this might cause them to contribute poorly to chimaeric conceptuses when aggregated with more precociously developing embryos is discussed.
